AFFY TERMINAL END-LABELLING
Affymetrix has produced a number of Genechip Tiling Arrays in recent years to accommodate researchers' endeavors to more fully interrogate genomes for multiple applications. For example, the Genechip Tiling Arrays have allowed researchers to do whole-genome screening of DNA binding proteins (ChIP-on Chip experiments) and full transcriptome mapping. Selection and amplification of targets for tiling array applications are usually highly specific and there is not a blanket assay designed for generating these targets. The CAT is now offering terminal end-labeling of targets that have been generated by researchers (and amplified using a combination of dTTP and dUTP) to hybridize to Tiling arrays. Because the end-labeling reaction is preceded by a Uracil DNA Glycosylase/APE 1 directed fragmentation, samples submitted must be double-stranded and have UTP incorporated at a specific concentration. This can be achieved via particular kits supplied from Affymetrix (for transcriptome mapping) or by following Affymetrix's recommended PCR procedure for amplifying targets for subsequent hybridization to Genechip Tiling Arrays.
